Effect of Composite of Bone Morphogenetic Protein and Plaster of Paris on Healing of Bone Defect in the Rat Tibia



Abstract
It is well established that the bone morphogenetic protein (BMP) has a strong bone forming activity in the experiments of dog, rat, rabbit and other mammals.
Although there had beem many investigations about carrier for BMP, which should be immunologically inert, easily resorbed, and provide appropriate mechanical stability, there was no general agreement about the best type of carrier.
To investigate the effectiveness of plaster of Paris(PP) as a carrier for BMP BMP/PP composite was inserted into the defect of the right proximal tibia of Sprague Dawley rat. Control PP without BMP was inserted into the left side.
The composite was made from 1mg of BMP, 5mg of PP and 5¥ìl of distilled water, and the control PP from 5mg of PP and the control PP from 5mg of PP and 5¥ìl of distilled water.
The control PP and BMP/PP composite did not cause foreign body reaction or inflammation and was substituted for new bone. The BMP/PP composite was resorbed early after implantation and produced more rapid bone formation. The amount of bone
formation
caused by BMP/PP composite was abundant enough to expand the proximal tibial metaphysis. On analysis of roentgenograms of the tibia, the AP and lateral diameters and the area of transverse cut surface was increased significantly from two weeks
after
implantation, in the BMP/PP composite group.
These results suggest that BMP/PP composite system is one of the most effective BMP carrier system.
